Stanford researchers have traced a plausible immune pathway that may explain why a very small number of people develop myocarditis after

mRNA COVID-19 vaccination. By comparing blood from affected and unaffected recipients, they found heightened activity of CXCL10

and interferon-gamma—immune signals that, in rare cases, appear capable of driving an inflammatory cascade in heart tissue. Lab models

showed macrophages and T cells amplifying this response, with cardiac markers of injury and impaired contraction emerging in both mouse hearts and human heart–like spheroids.

When scientists blocked CXCL10 and interferon-gamma, inflammation dropped while general immune protection largely remained,

hinting at future targeted therapies. Genistein, a soy-derived compound, showed anti-inflammatory effects in the lab but is far from a recommended treatment.

The work does not challenge the overall safety and benefits of vaccination; instead, it sharpens the focus on rare reactions, younger male vulnerability, and how to make next-generation mRNA therapies even safer through precise control of inflammatory pathways.
